Durastab™ series of BCIP/NBT membrane/precipitation substrates feature low background, high stability, sensitivity, and minimal batch-to-batch variation. The series provides blue and purple signal systems, along with BCIP/NBT substrates dedicated for immunohistochemistry (IHC). BCIP/NBT Purple One Component Membrane Substrate produces a dark purple signal and is suitable for applications including Western blot and microarray analysis.

Instruction:
Durastab™ BCIP/NBT Purple One Component Membrane Substrate is ready-to-use and should be equilibrated to the reaction temperature before application.
Chromogenic Reaction Procedure (Western Blot):
1. Wash: After alkaline phosphatase (AP) conjugate incubation, wash the membrane 3 times with TBST washing buffer.
2. Chromogenic Reaction: Add 1 mL substrate and incubate at 20–37 °C until bands appear dark purple.
3. Stop: Wash 2–3 times with 1.5 mL deionized water to terminate the reaction before background color develops.
4. Detection: Analyze bands by visual inspection or by imaging to quantify gray-scale values.

Q1: Which enzyme do BCIP/NBT substrates work with?
A: They are alkaline phosphatase (AP) substrates: AP dephosphorylates BCIP, which then precipitates with NBT as an intense blue-purple product on membranes and tissue sections.
Q2: What applications are BCIP/NBT substrates suited for?
A: Western blot, dot blot, membrane-based assays and immunohistochemistry - anywhere a permanent, scanner-detectable precipitate from an AP conjugate is required.
Q3: What is the difference between the BCIP/NBT versions?
A: BCIB (One/blue) is the standard single-component formulation; BCPE adds enhanced purple intensity; BCIS (Plus) is a higher-sensitivity version; BCFA (Fast) gives faster signal development for shorter protocols.
Q4: Do I need a reader to detect the signal?
A: No. The precipitated product is visible to the eye and is documented with a scanner or camera, which makes the substrates convenient for routine blot work.
Q5: Is there a drop-in alternative to my current BCIP/NBT substrate?
A: BCIP/NBT Purple One Component Membrane Substrate is a one-component AP substrate that produces the same blue-purple precipitate on membranes and tissue sections, so it can usually replace your current BCIP/NBT reagent without protocol changes. Match the version to your sensitivity and speed needs (standard, enhanced, plus or fast) and confirm spot intensity in a side-by-side blot - samples are available on request.
