Durablock™ Casein Blocking Solution is an optimized blocking system based on high‑purity chemically modified casein, delivering superior blocking performance for saturating polystyrene microplates or other solid surfaces. This effectively reduces background, lowers the coefficient of variation (CV), and enhances detection sensitivity.

Durablock™ Casein Blocking Solution is an optimized blocking system based on high‑purity chemically modified casein, delivering superior blocking performance for saturating polystyrene microplates or other solid surfaces. This effectively reduces background, lowers the coefficient of variation (CV), and enhances detection sensitivity. Upon incubation with the Casein Blocking Solution, unoccupied sites on the microplate or nitrocellulose membrane become saturated, preventing nonspecific binding of analytes or detection antibodies to the surface. This minimizes background and improves assay sensitivity. Based on chemically modified, high‑purity casein, this product significantly outperforms other commercial or in‑house casein solutions prepared by simple dissolution and filtration. For many assays, Casein Blocking Solution provides maximal saturation blocking and exceptionally low inter‑assay variation. The high‑purity, chemically modified casein molecules help minimize interactions between the blocking layer and the sample matrix. This results in both highly efficient blocking for more reliable detection outcomes and a reduction in cross‑reactivity while maximizing surface coverage.

Application:
ELISA：Ready‑to‑use for blocking after antibody or antigen coating.
Western blotting：Ready‑to‑use for blocking after membrane coating.
Instruction:
Durablock™ Casein Blocking Solution is ready‑to‑use. Allow it to equilibrate to the reaction temperature, then mix well before application.
Microplate Blocking Procedure (Example):
1.After antigen or antibody coating;
2.Wash: Remove unbound protein by washing twice with PBS/TRIS washing buffer (without surfactant);
3.Blocking: Add an appropriate amount of Durablock™ Casein Blocking Solution and incubate at 20–37 °C for 30–60 minutes;
4.Detection: Detection: Aspirate (or blot off) the Casein Blocking Solution and proceed directly to downstream detection; alternatively, wash away excess blocking solution twice with PBS/TRIS washing buffer (without surfactant) before performing downstream detection.
