Coating Buffer CB is used to dilute antigens or antibodies for coating or spotting onto polystyrene microplates, nitrocellulose membranes or other protein-binding surfaces, thereby immobilizing the antigen or antibody on the solid support.

Instruction:
Storage at 2-8 °C or freezing may cause salt crystallization. Before use, equilibrate to room temperature and shake thoroughly to dissolve all salt crystals. Dilute Coating Buffer 10× CB 10-fold with deionized water before use: add 1 part of Coating Buffer 10× CB to 9 parts of deionized water to obtain Coating Buffer 1× CB.
Typical coating antibody concentrations for standard ELISA range from 0.25 µg/mL to 2 µg/mL. The optimal coating concentration depends on the solid support and on the protein or antibody, and the optimal incubation time may vary. The coating process should therefore be optimized. For some proteins or antibodies, coating buffer at pH 9.6 performs better; for others, pH 7.4 is more suitable. For newly developed immunoassay protocols, we strongly recommend evaluating both Coating Buffer 10× CB and 10× PBS.
For nitrocellulose membrane coating, adding 3% trehalose to 1× coating buffer may improve coating performance.
