About UsTMB Blue One Component Membrane Substrate (TMMB) is a precipitating TMB substrate for membrane, blot and chip-based detection. It produces a dark blue precipitate and is suitable for applications such as Western blot and microarray analysis. Art no: TMMB Package size: 100mL, 1L, 5L, 10L, bulk pH value: 4.5±0.5 Shelf life: 3 years Storage: 2-8℃ Application: Immunoblot, Western blot, Northern blot, Microarray, ELISpot etc.
Tell us what you need and we will send the file to your email shortly.
Durastab™ series of membrane/ precipitation substrates features low background, high stability, sensitivity, and minimal batch-to-batch variation. The series provides blue, purple, and dark green signal systems for flexible selection in kit development and production. TMB Blue One Component Membrane Substrate produces a dark blue precipitate and is suitable for applications such as Western blot and microarray analysis.
Typical applications: Western blot, dot blot and membrane-based detection; the chip formulations extend the series to microarray and biosensor surfaces.
Instruction:
Durastab™ TMB Blue One Component Membrane Substrate is ready‑to‑use and should be equilibrated to the reaction temperature before use.
Chromogenic Reaction Procedure (Western Blot):
1. Wash:After HRP conjugate incubation, wash the membrane 3 times with PBST/TBST washing buffer;
2.Chromogenic Reaction: Add 1 mL substrate and incubate at 20–37 °C until bands appear dark blue;
3. Stop: Stop the reaction by washing 2–3 times with 1.5 mL deionized water before background develops;
4. Detection: Analyze bands by visual inspection or by imaging to quantify gray‑scale values.

Q1: What is the difference between TMB membrane substrates and TMB chip substrates?
A: Membrane formulations (TMMS, TMEN, TMMG, TMMB) are tuned for Western and dot blotting on nitrocellulose and PVDF, while chip formulations (TMEC, TMCG, TMCB, TMCP) deposit a tighter precipitate designed for microarray spots and biosensor surfaces.
Q2: Why are blue, green and purple versions available?
A: The precipitating substrates leave permanent, water-insoluble spots in different colors, so multiplex blots and arrays can distinguish several targets on one membrane without a chemiluminescence imager.
Q3: How do I stop the color development on a membrane?
A: Rinse the membrane with deionized water once the desired spot intensity is reached. The precipitated product is stable after drying and can be documented by scanner or camera.
Q4: Are these substrates supplied ready to use?
A: Yes. All membrane and chip formulations are one-component, HRP-triggered precipitating TMB substrates - no mixing step is required before use.
Q5: Can I switch from another precipitating TMB substrate to TMB Blue One Component Membrane Substrate?
A: Yes for standard membrane and chip workflows. TMB Blue One Component Membrane Substrate is a one-component, HRP-triggered precipitating formulation that develops permanent, water-insoluble spots on nitrocellulose, PVDF and array surfaces; stop with deionized water as usual and document by scanner or camera. For multiplex color layouts, pair it with the other color versions of the series.
