About UsTMB (3,3′,5,5′-tetramethylbenzidine) is the standard chromogenic substrate used with the HRP (horseradish peroxidase) detection enzyme in colorimetric ELISA and other HRP-based immunoassays. The signal-generation step of an immunoassay is critical to an accurate measurement, and choosing the right TMB substrate comes down to matching three assay-dependent properties: development speed (kinetic rate), sensitivity, and dynamic range. This guide explains how these trade off against one another and which Durastab™ formulation fits your workflow.
Durastab™ offers four one-component, ready-to-use TMB substrates for assay developers and kit manufacturers. They all feature low background, high stability and minimal lot-to-lot variation, and differ mainly in their kinetics and detection range — so you can choose the profile that most closely matches your immunoassay.

All Durastab™ TMB substrates are one-component, ready-to-use solutions — simply warm to room temperature and pipette. Because there is no separate component to premix, you eliminate mixing error, reduce handling time and avoid waste from unused A/B mixes, which helps streamline assay manufacturing.

you measure low-abundance analytes or want to detect weak positives with minimal signal loss. Its fast ramp yields strong signal quickly — just control the stop time.
you run routine batch ELISAs and want a dependable, middle-of-the-road substrate with no mixing.
your standard curve spans a wide concentration range and you want accurate quantification across it. The broader linear range reduces re-runs on out-of-range samples.
you need an ultra-slow substrate with the widest dynamic range — ideal for automation, high-throughput runs, or applications where a forgiving timing window matters.
Fast substrates (High Sensitive) save time and push signal higher, but color keeps climbing, so timing the stop is more critical. Slow substrates (Slow Plus) trade a little speed for a wide, forgiving endpoint and steadier automation behavior.
High sensitivity improves detection at low concentrations. Wide dynamic range extends the linear portion of the standard curve. Simply put: sensitivity helps you detect less; dynamic range helps you quantify more accurately across a spectrum.
Keep conditions consistent when comparing — same HRP concentration, substrate volume and temperature.
Mind platform temperature; development is faster at 37 °C than at room temperature.
Plan the endpoint read: read at 450 nm right after stopping, or at 650 nm if reading without stopping.
How do I choose a TMB substrate? Match speed, sensitivity and range to your assay. Low-concentration detection favors High Sensitive; wide-ranging samples favor Board Range or Slow Plus; routine work favors One Component.
Which is the most sensitive? High Sensitive develops signal fastest and is built for maximum sensitivity in low-analyte assays.
Do you offer a slow TMB? Yes — Slow Plus develops over roughly 15–30 min and gives a forgiving timing window.
Are all four ready-to-use? Yes. All are one-component, ready-to-use solutions stored at 2–8 °C, read at 450 nm after stopping (or 650 nm without stopping).
